Sapient’s ImmuneSeek is an innovative workflow for tumor immune profiling, enabling the direct measurement of immune cell states and signaling pathways in human tumors for precise characterization of immune activity and therapeutic response and resistance.
The success of modern oncology therapeutics increasingly depends on the functional state of the tumor immune microenvironment. Therapies such as immune checkpoint inhibitors, CAR-T, T-cell engagers, ADCs, and radioligand therapies all rely on complex interactions between tumor cells and immune populations – yet these critical properties, governed by the functional tumor immune proteome, are rarely captured by conventional genomic approaches.
We developed ImmuneSeek to address this challenge by directly quantifying and mapping immune biology in human tumors using a mass spectrometry-based discovery proteomics workflow.
What dimensions of tumor immune biology does ImmuneSeek profile?
ImmuneSeek shifts tumor immune profiling from inference to direct measurement of functional pathway activity, delivering quantitative insight into immune biology that determines therapeutic response. This includes measurement of:
- Immune cell composition and infiltration
- Functional activation of immune pathways
- Antigen presentation capacity
- Immune suppression and checkpoint signaling
- Tumor–immune interaction dynamics
Importantly, ImmuneSeek is optimized for both fresh-frozen and FFPE human tumor samples, enabling quantitative characterization of immune function in archived samples to deliver novel insights without requiring new biopsies.
What new insights can I gain from directly measuring the proteins that govern immune biology?
ImmuneSeek applies deep discovery proteomics to comprehensively profile and map immune biology across multiple dimensions at the protein level, revealing functional immune activity rather than inferred potential.
This includes direct quantitation of proteins that define immune cell populations, cytotoxic effector programs, T-cell exhaustion and regulatory states, and myeloid and macrophage polarization. It also captures activation of key immune pathways, such as interferon and cytokine signaling, and quantifies resistance through the measurement of protein-level adaptations such as loss of antigen presentation and interferon signaling defects.

What development questions can I answer using ImmuneSeek for tumor immune profiling?
Precise characterization of immune activity, response, and resistance is essential to the development of modern oncology therapeutics. ImmuneSeek delivers this depth of insight to optimize patient selection, biomarker development, and therapeutic strategy, allowing teams to answer questions such as:
| Development Question | ImmuneSeek Insight |
|---|---|
| Are immune pathways functionally active? | Direct measurement of pathway activation |
| Why are patients not responding to immunotherapy? | Identification of immune resistance mechanisms |
| How can patients be stratified? | Functional immune state classification |
| Which combinations should be pursued? | Identification of suppressive or compensatory pathways |
ImmuneSeek is offered as a standalone workflow or within Sapient’s broader Tumor Protein Mapping Platform which also includes SurfaceSeek™ to identify druggable tumor cell surface proteins, SignalingSeek™ to measure signaling pathway activation, and ResistanceSeek™ to map resistance biology – directly in fresh-frozen and FFPE human tumors.
Move from descriptive immune biology to actionable insight with ImmuneSeek – get started today
With this workflow, we empower drug developers with the insights needed to determine whether immune cells are functionally active, to map which pathways are driving response or suppression, and to decipher why patients succeed or fail on therapy.
