What molecular signals can we find via large-scale metabolomics analysis across thousands of people at a time? And what can they tell us about the dynamics of health, disease, and drug response to inform drug development and better diagnostics?

In this Omics Brief, we provide a short review of Sapient’s recently published study, “Rapid Liquid Chromatography-Mass Spectrometry (rLC-MS) for Deep Metabolomics Analysis of Population Scale Studies” to highlight the technical innovations that have enabled one of the largest non-targeted metabolomics analyses conducted to date – and key discoveries made, including a dynamic metabolic aging clock to predict accelerated aging.

The Omics Brief highlights:

  • Key innovations enabling the novel rLC-MS method for generating large-scale, non-targeted metabolomics data and making it readily actionable, including machine learning (ML)-based spectral handling and AI-enabled metabolite identification.
  • Important findings from rLC-MS metabolomics analysis of 26,042 plasma samples acquired across multiple timepoints from 6,935 individuals which reveal a varied chemical landscape across human populations.
  • The discovery of unique “metabotypes”, or metabolic phenotypes, showing clearly distinguishable subgroups to predict metabolic disorders independent of traditional disease risk factors.
  • The development of an ML-based metabolic aging clock model trained on large-scale rLC-MS data that captures dynamic changes in biological aging, including ‘reversal’ of accelerated aging following definitive treatment like organ transplantation.
